ICOS Binding to B7-H2-transfected HEK293 Cell Line is Blocked by Mouse B7-H2 Antibody.

In a functional flow cytometry test, Recombinant Mouse ICOS Fc Chimera (168-CS; 50 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with mouse B7-H2. Binding is blocked by (A) 2.5 µg/mL of Rat Anti-Mouse B7-H2 Monoclonal Antibody (Catalog # MAB1582) but not by (B) Rat IgG1 Isotype Control (MAB005) at 2.5 µg/mL. Protein binding was detected with APC-conjugated anti-Human IgG Fc Monoclonal Antibody (FAB110A).

Background: B7-H2

B7-H2, also called B7RP-1 and ICOSL, is a 60 kDa member of the B7 family of immune costimulatory proteins. It is expressed on resting B cells, dendritic cells, monocytes, alveolar epithelial cells, and placental trophoblasts. B7-H2 interactions with ICOS promote lymphocyte differentiation and activation. Within the ECD, mouse B7-H2 shares 49% and 70% aa sequence identity with human and rat B7-H2, respectively.
