Detection of Human Livin by Western Blot.
Western blot shows lysates of SK-Mel-28 human malignant melanoma cell line. PVDF membrane was probed with 0.5 µg/mL Goat Anti-Human Livin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1161) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). For additional reference, Recombinant Human Livin a/KIAP (Catalog # 1161-LV) and Recombinant Human Livin beta/ML-IAP (Catalog # 787-LV) (0.5 ng/lane) were included. A specific band for Livin was detected at approximately 34 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 4.Detection of Human Livin by Simple WesternTM.
Simple Western lane view shows lysates of SK-Mel-28 human malignant melanoma cell line, loaded at 0.2 mg/mL. A specific band was detected for Livin at approximately 42 kDa (as indicated) using 5 µg/mL of Goat Anti-Human Livin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1161) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). Background: Livin
Livin alpha (KIAP) and Livin beta (ML-IAP) are two splice forms generated from the livin gene. Livin is a member of the inhibitor of apoptosis protein (IAP) family that contains a baculovirus single IAP repeat (BIR) and a C-terminal RING finger domain. Livin inhibits the activation of caspases 9 and/or 3 when activated in specific cell lines. Livin alpha is a splice variant 18 amino acids longer in the BIR-RING linking region than the livin beta splice variant. The isoforms differ in their apoptotic properties.
