Activity

Measured in a cell proliferation assay using human CD3+ T cells in the presence of anti-CD3. Kanner, S.B. et al. (1992) J. Immunol. 148:2023.
The ED50 for this effect is 1-4 µg/mL in the presence of anti-CD3 immobilized at 20 ng/mL.
For carrier-free vials, the Recombinant Human CD58/LFA‑3 (rhCD58) is directly coated to plates. For vials containing carrier, plates are coated with 5 µg/mL Mouse Anti-polyHistidine Monoclonal Antibody (Catalog # MAB050) before rhCD58 is added.

What does CF mean?

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?

In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

Reconstitution Reconstitute at 100 μg/mL in sterile PBS containing at least 0.1% human or bovine serum albumin.

Background: CD58/LFA-3

T cells require a signal induced by the engagement of the T cell receptor and a “co-stimulatory” signal(s) through distinct T cell surface molecules for optimal T cell expansion and activation. Many cell-bound receptor-ligand pairs have now been shown to be involved in T cell co-stimulation including CD58/CD2 in humans and CD48/CD2 in mice and rats. CD58, also known as lymphocyte function-associated antigen (LFA-3), is a 210 amino acid protein that belongs to the CD2 family of the immunoglobulin superfamily (1). CD58 is widely expressed on hematopoietic and non-hematopoietic human tissue and has been found on leukocytes, erythrocytes, endothelial cells, epithelial cells and fibroblasts of human origin (2). No mouse or rat homolog of CD58 has as of yet been identified. CD58 has only one known ligand, CD2. CD2 is expressed on T cells, NK cells and dendritic cells (2‑4). CD2 ligation by CD58 has been shown to mediate T cell adhesion, T cell activation, T cell cytokine production and T cell and NK cells cytotoxic activity (1, 3, 5, 6). In dendritic cells, CD2 engagement increases MHC Class II, CD40, CD80, CD86, CD58 and CCR7 and induces IL-1 beta and IL-12 cytokine secretion (4).

  • Q: This protein datasheet indicates I need to use a cross-linking antibody, Catalog # MAB050, for biological activity. What is this antibody and is it really necessary?

    A: The antibody is directed against a 6x histidine repeat and is recommended for use as a cross-linker of proteins with 6x his-tag. Crosslinking is often used for proteins that require receptor trimerization and can result greater biological activity. R&D Systems Quality Control tests the performance of these proteins in the presence of the cross-linking antibody. Therefore, it is necessary to use this antibody when trying to achieve the same level of specific activity described in the datasheet.
