Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32470) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: IL-23 Antibody (JE38-99) [NBP3-32470] - Immunohistochemical analysis of paraffin-embedded human lymph nodes tissue with Rabbit anti-IL-23 antibody (NBP3-32470) at 1/400 dilution.The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32470) at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: IL-23
IL-23 (Interleukin-23) is a disulfide-linked cytokine composed of a p19 subunit that is unique to IL-23 and a p40 subunit that is shared with IL-12. It is produced by activated macrophages, microglia, and monocyte-derived dendritic cells in response to pathogens. IL-23 induces the earliest recruitment of neutrophils to the site of infection and promotes the development and maintenance of Th17 cells. It also enhances the development of Th17 mediated autoimmunity and tumor progression. IL-23 signals through a receptor complex consisting of IL-12 R beta 1 and IL-23 R. This complex is expressed in mouse Th1 and Th2 cells, bone marrow dendritic cells, activated macrophages and CD4+ CD45Rb(low) memory T cells.
