TNF RI/TNFRSF1A Inhibition of TNF‑ alpha-induced Cytotoxicity and Neutralization by Human TNF RI/TNFRSF1A Antibody.
Recombinant Human TNF RI/TNFRSF1A (Catalog # 636-R1) inhibits Recombinant Human TNF-a (Catalog # 210-TA) induced cytotoxicity in the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line). Inhibition of Recombinant Human TNF-a (0.25 ng/mL) activity elicited by Recombinant Human TNF RI/TNFRSF1A (0.3 µg/mL) is neutralized (green line) by increasing concentrations of Human TNF RI/TNFRSF1A Monoclonal Antibody (Catalog # MAB625). Inhibition assays with antibody against tumor necrosis factor-receptor 1 (TNFR1AB). (A) Analysis of nuclear factor-kappa-B (NF-kappa B) phosphorylation upon blockade of TNFR1. Dental pulp stems (DPCs) were incubated with different concentrations (0, 4, 6, and 10 μg/mL) of TNFR1AB for 1 hour prior to treatment with TNF-alpha (10 ng/mL). Phosphorylation of NF-kappa B (p65, p105) was examined by Western blot at 5 minutes after stimulation. TNFR1AB inhibited TNF-alpha -induced phosphorylation of p105 and p65 in a dose-dependent manner, and a maximum effect was observed with a concentration of 10 μg/mL. (B) Flow cytometric analysis of expression of stage-specific embryonic antigen 4 (SSEA-4). TNFR1AB only partially inhibited TNF-alpha -induced increase in the number of SSEA-4+ cells. (C) Analysis of mRNA levels of OCT-4 and NANOG upon inhibition of TNFR1. Although TNF-alpha treatment increased the gene expression of OCT-4 and NANOG, TNFR1AB nullified the TNF-alpha -induced increase OCT-4 mRNA levels only. **P <0.01, one-way analysis of variance (ANOVA), Tukey test. Neutralization
                          Measured by its ability to neutralize TNF RI/TNFRSF1A-mediated inhibition of cytotoxicity in the L‑929 mouse fibroblast cell line. The Neutralization Dose (ND50) is typically 1-6 µg/mL in the presence of 0.3  μg/mL Recombinant Human TNF RI/TNFRSF1A, 0.25 ng/mL Recombinant Human TNF‑ alpha, and 1 µg/mL actinomycin D.
