Detection of E6AP/UBE3A in Human Liver.

E6AP/UBE3A was detected in immersion fixed paraffin-embedded sections of human liver using Mouse Anti-Human E6AP/UBE3A Monoclonal Antibody (Catalog # MAB11546) at 5 µg/ml for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001) or the HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the nucleus and cytoplasm. Detection of E6AP/UBE3A in A172 Human Cell Line.

E6AP/UBE3A was detected in fixed A172 human glioblastoma cell line using Mouse Anti-Human E6AP/UBE3A Monoclonal Antibody (Catalog # MAB11546) at 8 µg/ml for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Background: E6AP/UBE3A

Ubiquitin-protein ligase E3A, also known as E6AP, is an E3 ligase that accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and transfers it to its substrates. Several key substrates for UBE3A have been identified, including BMAL1, the PML tumor suppressor, PGR, and p53/TP53 suggesting a role for UBE3A in regulation of the circadian clock, tumor regulation, transcriptional coactivation of the progesterone receptors, and regulation of neoplastic progression of cells infected by high-risk human papilloma virus (1-4). Defects in activity are linked to Angelman syndrome, a neurodevelopmental disorder, as well as autism spectrum disorders implicating a role for UBE3A in regulation of neurobiological functions (3, 5, 6).
