Product Description
Cell Counting Kit-8 is a viability and proliferation assay test solution. It is a supplied as a ready-to-use 1-bottle solution (5 mL; 500 tests) and enables the colorimetric determination of the number of viable cells in cell proliferation and cytotoxicity assays. Cell Counting Kit-8 utilizes a highly water-soluble tetrazolium salt (WST-8), which is reduced by cellular dehydrogenases to an orange formazan dye. The color change can be detected in the 430 - 490 nm range and Cell Counting Kit-8 exhibits improved sensitivity compared with other tetrazolium salt-based assays such as MTT, XTT or MTS. Cell Number Determination.
HeLa cells 1x104 cells/well in MEM were incubated overnight (37°C, 5% CO2) in clear 96-well plates. 10 μL Cell Counting Kit-8 or 10 μL MTT (Catalog # 5224) was added and incubated for a further 90 mins. The absorbance was measured at 450 nm or 570 nm, higher sensitivity was achieved via the Cell Counting Kit-8 assay.Cytotoxicity Assay.
