Summary
Performance Comparison: Single Plex vs. Parallel 4-Plex Workflows
In this app note using both single plex and parallel 4-plex workflows for SPEAR UltraDetect pTau 217, pTau 231, NF-L and GFAP Assays, equivalent performance was demonstrated. Across the single plex and parallel plex studies, the parallel 4-plex workflow produced quantitative results in strong agreement with the single plex format, exhibiting comparable precision and fold separation between disease and healthy control groups.
Biomarkers Evaluated (pTau 217, pTau 231, NF-L, GFAP)
Using this Parallel Plex Solution researchers can measure multiple biomarkers in parallel—each with the single plex-grade sensitivity, specificity, and precision—while consuming a single small-volume sample aliquot.
Highlights:
- Demonstrates analytical equivalency between single plex and parallel 4-plex SPEAR UltraDetect workflows.
- Enables simultaneous measurement of pTau 217, pTau 231, NF-L, and GFAP from a single small-volume sample aliquot.
- Maintains single-plex–grade sensitivity, specificity, and precision in a multiplexed format.
- Enables concurrent analysis of closely related biomarkers, such as multiple Tau or pTau species, that cannot be measured together using conventional multiplex immunoassays.
- Supports the use of SPEAR UltraDetect parallel plex workflow as an efficient and scalable solution for multi-biomarker analysis in research applications.
