Detection of Rat IgG Primary Antibody by Immunocytochemistry

Detection of human Vimentin by incubating cells with 8 ug/mL of Rat anti-Human Vimentin Biotinylated monoclonal antibody (BAM2105) at room temperature for 1 hour followed by incubation with 1:5000 dilution of NorthernLights™ Streptavidin NL637-conjugated secondary antibody for 1 hour at room temperature. Note strong positive staining in A172 cells (left panel, positive control) and a lack of non-specific background staining in the absence of primary antibodies (right panel, negative control).

Background: Streptavidin

Streptavidin is a tetrameric protein purified from Streptomyces sp. that binds very tightly to the vitamin biotin with a Kd of ~ 10-14 mol/l. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.
