Biological Activity for NMS 873

NMS 873 is a potent p97 ATPase (VCP) allosteric inhibitor (IC₅₀ = 30 nM). Selective over all other AAA ATPases, HSP90 and kinases tested (IC₅₀ >10 μM). Activates the unfolded protein response (UPR) and modulates autophagosome maturation. Exhibits antiproliferative activity in cancer cells in vitro (IC₅₀ values are 400 and 700 nM for HCT116 and HeLa cells respectively). Reduces p97-sensitization to trypsin digestion.

Used NMS at 30 nM and 300 nM in Dictyostelium, treated for 1 hour or 24 hours. Then mTORC1 activity was monitored using a western blot for p-4E-BP1. No significant change in p-4E-BP1 levels was observed
