Key information: Janelia Fluor® 646 Tre is a red fluorescent probe for imaging Mycobacterium tuberculosis, after metabolic incorporation of the trehalose component in the lipid-rich cell envelope.Used for: imaging Mycobacterium tuberculosis in TB drug susceptibility testing.
Application: fluorescent microscopy and flow cytometry.
Properties and Photophysical Data: Excitation and emission maxima (λ) are 646 nm and 664 nm, respectively; quantum yield = 0.54; extinction coefficient = 152,000 M-1cm-1 (measured in ethanol plus 0.1% TFA); A280 correction factor = 0.19; lactone-zwitterion equilibrium constant (KL-Z) = 0.0012.

Application of Janelia Fluor® 646 Tre for live bacterial imaging.
Msmeg cells were incubated 4-hour at 37 ºC in the presence of 10 µM Janelia Fluor® 646 Tre. Cells were washed in DPBS and imaged using a Nikon A1R Laser Scanning Confocal Microscopy. Data kindly supplied by the Swarts lab, Central Michigan University.Application of Janelia Fluor® 646 tre for flow cytometry using APC channel.
Msmeg cells were incubated for 4 hours and Mtb cells for 16 hours at 37 ºC in the presence of 10 µM Janelia Fluor® 646 Tre. Flow cytometry was performed on a BD Biosciences C6 Accuri™ Plus flow cytometer instrument equipped with a 640-nm red excitation and APC (675/25 nm) emission filter.
