The Proteome Profiler Human Cytokine Array Kit, Panel A is a membrane-based sandwich immunoassay. Samples are mixed with a cocktail of biotinylated detection antibodies (Step 1) and then incubated with the array membrane which is spotted in duplicate with capture antibodies to specific target proteins (Step 2). Captured proteins are visualized using chemiluminescent detection reagents (Step 3). The signal produced is proportional to the amount of analyte bound.
Why Use an Antibody Array to Detect Multiple Cytokines?
Determining the expression of multiple cytokines in a single sample can be expensive, time consuming and can require specialized equipment. Performing multiple immunoprecipitations and Western blots requires time, labor, and reagents. The use of a multiplex antibody array to detect multiple cytokines in a single sample can be cost-effective and also save time and sample.

Data Examples
Detection of Multiple Human Cytokines in Cell Culture Supernates. Cells were treated as described below. Following treatment, cell culture supernates were analyzed using the Proteome Profiler Human Cytokine Array Kit, Panel A. Human peripheral blood mononuclear cells were treated with phytohemagglutinin (PHA) for 24 hours. B. TF-1 human erythroleukemia cells were treated with phorbol 12-myristate 13-acetate (PMA) for 24 hours. C. THP-1 human acute monocytic leukemia cells were treated with lipopolysaccharide (LPS) for 16 hours.
Detection of Multiple Cytokines in Human Serum. Human serum was collected from four donors (A – D), and the relative levels of multiple cytokines were determined using the Proteome Profiler Human Cytokine Array Kit, Panel A.
Detection of Multiple Cytokines in Human Tissues and Cell Lysates. Extracts were generated from human tissue or cell lines indicated below and analyzed for relative cytokine expression using the Proteome Profiler Human Cytokine Array Kit, Panel A. A. Normal human lung tissue. B. Malignant human lung cancer tissue. C. Human A431 epithelial carcinoma cells.
