Thymidylate Synthase Antibody was purified from rabbit serum after repeated immunizations with recombinant human Thymidylate Synthase (36 kDa) produced in E.coli. (Uniprot: P04818)

Reactivity Notes

In general, this antibody can detect antigen in a variety of human cells and tissues, as well as bacteria, African green monkey, rat and mouse. Somewhat lower dilutions may be required in some non-human cell lines.

Specificity

Anti-THYMIDYLATE SYNTHASE IgG fraction is directed against human Thymidylate Synthase and is useful in determining its presence in various assays. Because inhibition of Thymidylate Synthase prevents DNA synthesis and cell proliferation, the enzyme is an important target for cancer chemotherapeutic drugs, specifically the fluoropyrimidine group of antineoplastic drugs used to treat solid tumors. In general, this antibody can detect antigen in a variety of human cells and tissues, as well as bacteria, African green monkey, rat and mouse. Somewhat lower dilutions may be required in some non-human cell lines. Anti-Thymidylate Synthase can detect Thymidylate Synthase by immunochemistry in proliferating cell cultures and tissues but does not stain nonproliferating cells. Normal colon mucosa shows weak staining; however, some colorectal cancer specimens show very strong staining.

Background: Thymidylate Synthase

Thymidylate synthase (TS) is an intracellular enzyme critical for de novo synthesis of DNA. In cancer, expression of TS is often elevated and becomes further elevated as a result of treatment with the most commonly used chemotherapeutic, 5-fluorouracil (5-FU). Resistance or lack of response to 5-FU is attributed to the elevation of TS activity. Overexpression of TS has been demonstrated in many types of tumors including breast, colon, gastric and melanoma and is usually predictive of shortened survival time.
