Specificity
Detects human Smad8 in direct ELISAs and Western blots. In direct ELISAs, approximately 90% cross-reactivity with recombinant (rh) Smad1 is observed, approximately 20% cross-reactivity with rhSmad4 is observed, and less than 1% cross-reactivity with rhSmad5, rhSmad7, and rhDNAK is observed.

Description
CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. Background: Smad8
Smads are a family of intracellular proteins that mediate signaling by members of the TGF-beta superfamily. Smads are divided into three distinct subgroups based on their different roles in TGF-beta family signal transduction: R-Smads (receptor-regulated), Co-Smads (common partner), and I-Smads (inhibitory). In general, activation of TGF-beta superfamily serine/threonine kinase receptors leads to the phosphorylation of R-Smads (Smads1, 2, 3, 5, and 8). This is followed by recruitment of the Co-Smad (Smad-4) into an R-Smad/Co-Smad complex that translocates to the nucleus to regulate gene expression. I-Smads (Smad6 and Smad7) antagonize this signaling pathway by preventing R-Smads from interacting with the receptor, or by competing with Co-Smads for the generation of R-Smad/Co-Smad complexes.
