Background: Pichia Pastoris

Pichia pastoris, a methylotrophic yeast, has been used as a heterologous protein expression system. It combines many of the benefits of E. coli expression with the advantages of expression in an eukaryotic system such as protein processing, folding, and posttranslational modifications. Pichia pastoris is capable of metabolizing methanol as the sole carbon source. The first step in the metabolism of methanol is the oxidation of methanol to formaldehyde by the enzyme, alcohol oxidase. Expression of this enzyme, coded for by the AOX1 gene, is tightly regulated and induced by methanol to very high levels: typically >30% of the total soluble protein in the cells.
