Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-FKBP15 antibody NB100-422 (lot 2) used for IP at 6 ug per reaction. FKBP15 was also immunoprecipitated by a previous lot of this antibody (lot 1) and rabbit anti-FKBP15 antibody NB100-423. For blotting immunoprecipitated FKBP15, NB100-422 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: FKBP15

Immunophilins are a family of soluble receptors capable of binding to one of two major immunosuppressant agents; cyclosporin A (CsA) or FK506. Proteins that bind FK506 are termed FK506 Binding Proteins (FKBPs) and those that bind cyclosporin A are called cyclophilins (CyP). Immunophilins function as cis-trans peptidyl-prolyl isomerases (PPIase) whose activity is inhibited by their respective immunosuppressant compounds. Thus, immunophilins accelerate folding of some proteins both in vivo and in vitro by catalyzing slow steps in the initial folding and rearrangement of proline-containing proteins.
