Description

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Alkaline Phosphatase [E.coli]

Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Western Blot: Alkaline Phosphatase, Bacterial Antibody [Biotin] [NB600-500] - Lane 1: Alkaline Phosphatase (E. Coli). Load: 50 ng per lane. Primary antibody: Rabbit anti-Alkaline Phosphatase (E. Coli) Antibody at 1:1,000 overnight at 4C. Secondary antibody: Peroxidase Rabbit secondary antibody at 1:40,000 for 30 min at RT. Block: incubated with blocking buffer for 30 min at RT. Predicted/Observed size: 50 kDa, 50 kDa for Alkaline Phosphatase (E. Coli).

Application Notes
This product has been tested against 1.0 ug of Alkaline Phosphatase [E.coli] in a standard ELISA using Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] (Goat) and (ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:25,000 to 1:125,000 of the reconstitution concentration is suggested for this product. A 1:10,000 dilution has been reported to be useful for the detection by immunoblot of an integral membrane protein from a gram-positive organism expressed as a LacZ/PhoA chimeric derivative in E.coli. The researcher should determine optimal titers for other applications.

Background: Alkaline Phosphatase, Bacterial

Bacterial alkaline phosphatase (also known as PhoA) is derived from E. coli. It appears to require export to the periplasm to show enzymatic activity. Fusion of the secreted alkaline phosphatase to an integral cytoplasmic membrane protein of E. coli shows different activities depending on where with the membrane protein the alkaline phosphatase is fused. Consequently, analysis of alkaline phosphatase fusions to membrane proteins of unknown structure are generally useful in determining their membrane topologies.
