Discover a novel workflow that streamlines charge–size variant characterization for complex antibody modalities. Dr. Srinivasa Rao (Bio-Techne) and Dr. Nina Jajcanin Jozic (Refeyn) illustrate how imaged capillary isoelectric focusing (icIEF) fractionation and mass photometry can be combined for deeper characterization of charge variants.

Utilizing the MauriceFlex™ System and Refeyn’s TwoMP mass photometer, they demonstrate a fast, simple application that utilizes charge variant separation and fractionation  followed by  downstream charge–size variant analysis to uncover differences between a bispecific antibody (Mosunetuzumab-axgb) and a research-grade biosimilar. The speakers demonstrate that icIEF-fractionation paired with label free mass photometry enables the direct tracking of LMWS and HMWS across acidic, main, and basic peaks under control and forced degradation conditions requiring only nanogram level material.

These results provide insights into bispecific charge variants and aggregation based on different conditions. These two compact, user-friendly instruments present a straightforward and time-saving workflow for the further characterization of charge variants in biomolecules.
