Immunocytochemistry/Immunofluorescence: TrkA Antibody (MNAC13) [NBP2-81074] - Immunofluoresence staining of fixed HeLa cells with anti-TrkA antibody (MNAC13) [NBP2-81074]. Immunofluorescence analysis of paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of MNAC13 [NBP2-81074] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1 ug/ml), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81074, DAPI, merged channels and a negative control. Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. This antibody binds to human TrkA, a receptor tyrosine kinase involved in the development and the maturation of the CNS and the PNS through regulation of proliferation, differentiation, and survival of the sympathetic and nervous neurons. It is a receptor for NGF and phosphorylates several downstream proteins regulating a variety of signalling cascades. This antibody neutralizes the NGF-TrkA interation.

Background: TrkA

The Trk proto-oncogene encodes a 140 kDa, membrane-spanning protein tyrosine kinase that is expressed only in neural tissues. Nerve growth factor (NGF) stimulates phosphorylation of Trk A in neural cell lines and in embryonic dorsal root ganglia. Affinity cross-linking and equilibrium binding experiments with 125I-labeled NGF indicate that Trk A binds NGF specifically in cultured cells with a dissociation constant of 10(-9) molar. The identification of Trk A as an NGF receptor indicates that this protein participates in the primary signal transduction mechanism of NGF (1). Trk A was found to be expressed in the nervous system and phosphorylated in response to NGF (Nerve Growth Factor). Somatic rearrangement(s) of the TRKA gene (also designated NTRK1) are responsible for formation of some oncogenes (2). Trk A is expressed in neural and nonneuronal tissues. Like RET, Trk A is often activated by rearrangements that involve one of at least five other genes in papillary thyroid carcinoma (PTC) (3).
