Localization

Western Blot: TDP-43/TARDBP Antibody (3H8) [NBP1-92695] - Analysis of whole brain lysates and nuclear inreached extract from whole brain using mouse mAb to TDP43, NBP1-92695, dilution 1:2,000 in green: [1] protein standard (red), [2] rat brain, [3] rat brain nuclear extract, [4] mouse brain, [5] mouse brain nuclear extract. Strong band at 43 kDa mark corresponds to TDP43 protein.
Immunocytochemistry/Immunofluorescence: TARDBP Antibody (3H8) [NBP1-92695] - NBP1-92695 was used to stain a section of formalin fixed adult rat brain, specifically the hippocampus. Hippocampal neuron nuclei are stained strongly. Chicken antibody to GFAP (NBP1-05198, green) shows the processes of astrocytic glial cells. Nuclei of all cells are revealed with DAPI DNA stain (blue). The TARDBP (3H8) antibody stains neuronal nuclei strongly and the nuclei of some non-neuronal cells much more weakly. Neuronal nuclei therefore look crimson, since they are both red due to the content of TARDBP and blue due to their content of DNA, stained blue with DAPI.
Immunohistochemistry Free-Floating: TDP-43/TARDBP Antibody (3H8) [NBP1-92695] - Analysis of rat hippocampus section stained with mouse mAb to TDP43, NBP1-92695, dilution 1:2,000 in red, and costained with chicken pAb to GFAP,, dilution 1:5,000 in green. The blue is DAPI staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. The TDP43 protein is concentrated in the nuclei of hippocampal neurons, while the GFAP antibody stains the intermediate filament network of astroglial cells.

Background: TDP-43/TARDBP

TARDBP (tar-DNA binding protein of 43) is a nucleic acids binding protein that regulates the process of transcription and splicing. Localized mainly in the nucleus, TARDBP is ubiquitously expressed with higher expression levels in pancreas, placenta, lung, genital tract and spleen, and has been suggested to implicate in microRNA biogenesis, cellular apoptosis as well as proliferation. TARDBP controls CFTR splicing and promotes CFTR exon 9 skipping by associating with UG repeated motifs in polymorphic region near 3'-splice site of this exon leading to aberrant splicing linked to pathological features typical of cystic fibrosis. TARDBP can repress HIV-1 transcription by binding to the HIV-1 long terminal repeat. Moreover, it stabilizes the low molecular weight neurofilament mRNA through a direct interaction with the 3' UTR. TARDBP was originally identified as a major protein component of the ubiquitin immuno-positive inclusions which are the pathologic substrate of common forms of ALS (amyotrophic lateral sclerosis) and FTD (frontotemporal dementia), and later, it was found that TARDBP gene mutations are inherited in an autosomal dominant manner in familial/sporadic ALS. TARDBP has also been involved in other neurodegenerative such as Alzheimer's and Parkinson's disease.
