This SREBP1 (clone 2A4) antibody is useful for WB where a band can be seen at 125 kDa (precursor) and additional bands may be seen at 60-70 kDa (cleaved).

In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue. The use of this antibody in IHC paraffin embedded tissue has been questionable.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: SREBP1

SREBP1 (sterol regulatory element binding protein 1, also called SREBF1 or BHLHD1) is a bHLH-LZ (basic helix-loop-helix-leucine zipper) transcription factor that acts as a key lipogenic transcription factor controlled by glucose/insulin and preferentially regulates lipogenesis via activating genes involved in fatty acid as well as triglyceride synthesis. It is a transcriptional activator required for lipid homeostasis and regulates transcription of the LDL receptor gene as well as the fatty acid and to a lesser degree the cholesterol synthesis pathway. SREBP1 has a dual sequence specificity binding to both an E-box motif (5'-ATCACGTGA-3') and to SRE-1 (sterol regulatory element 1; 5'-ATCACCCCAC-3'). It exists as multi-pass membrane protein in the membranes of ER, Golgi apparatus and cytoplasmic vesicle /COPII-coated vesicles and moves from ER to Golgi in the absence of sterols. It forms a tight complex with SCAP in ER membranes and interacts with LMNA. AMPK phosphorylate SREBP1 leading to suppression of protein processing and nuclear translocation, and target gene expression repression. Moreover, SIK1 mediated phosphorylation at Ser-402 site represses its activity through inhibition of DNA-binding.

Western Blot Protocol

1. Perform SDS-PAGE on samples to be analyzed, loading 20 ug of total protein per lane.
2. Transfer proteins to membrane according to the instructions provided by the manufacturer of the membrane and transfer apparatus.
3. Stain according to standard Ponceau S procedure (or similar product) to assess transfer success, and mark molecular weight standards where appropriate.
4. Rinse the blot.
5. Block the membrane using standard blocking buffer for at least 1 hour.
6. Wash the membrane in wash buffer three times for 10 minutes each.
7. Dilute primary antibody in blocking buffer and incubate overnight at 4C.
8. Wash the membrane in wash buffer three times for 10 minutes each.
9. Apply the diluted HRP conjugated secondary antibody in blocking buffer (as per manufacturers instructions) and incubate 1 hour at room temperature.
10. Wash the blot in wash buffer three times for 10 minutes each (this step can be repeated as required to reduce background).
11. Apply the detection reagent of choice in accordance with the manufacturers instructions.

**Note: Tween-20 can be added to the blocking or antibody dilution buffer at a final concentration of 0.05-0.2%.

  • Q: I would like to order SREBP-1 antibody, catalog number NB600-582. I am wondering if it might cross react with MDCK-C cells from dog?

    A: We have not tested NB600-582 with any dog samples, and cannot guarantee that it will cross-react. However, if you are interested in trying this antibody with dog samples you would be eligible for our Innovators Reward Program.

  • Q: I am looking for SREBF1 antibodies that can cross-react with Zebrafish srebf1. I am wondering whether you have any SREBF1 antibodies that have been tested for zebra fish or have high sequence identity in the immunogen region?

    A: Unfortunately we do not have any antibodies that have been tested in zebra fish. I have included the immunogen sequence for the antibodies it might be worth considering. I am not sure exactly what the zebra fish sequence is, I was unable to find it on uniprot.org, so I was unable to check the homology. The following antibodies all have available sample sizes.
