The original antibody was generated by immunizing 4-6 weeks old female ICR mice with recombinant protein comprising amino acids 488-590 of cytokeratin 5 in equal volume of Freund's incomplete adjuvant. (Uniprot# P13647)
Specificity
This antibody is capable of binding human cytokeratin 5 and human cytokeratin 6. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. Cytokeratin 5 is often expressed in a basal cell layer of normal mammary ducts. Studies have also shown that CK5/6 can be used as a characteristic marker of basal-like breast cancer.

Western Blot: Cytokeratin 5/6 Antibody (AbM58019-7H4) - Chimeric - BSA Free [NBP3-43564] - A431 cells lysate (35µg protein in RIPA buffer) was resolved via SDS-PAGE, and the subsequent blot was probed with the mouse version of NBP3-43564 at 0.01µg/ml before detection using an anti-mouse secondary antibody. A primary incubation of 1 hour was used, and proteins were detected by chemiluminescence.

The nuclear stain is DAPI (blue). Panels show, from left-right, top-bottom, NBP3-43564, DAPI, merged channels, and an isotype control.
