Quantifying biomarkers in biological fluids is increasingly vital for diagnosing, predicting, and monitoring neurodegenerative diseases. While cerebrospinal fluid (CSF) analysis has traditionally offered valuable insights, there is growing interest in blood-based biomarker detection due to its accessibility and minimally invasive nature. However, the significantly lower concentrations of these biomarkers in blood require highly sensitive and robust analytical platforms.
We present enhancements to the Simple Plex™ platform using the Ella instrument, an automated benchtop bioanalyzer, for the reliable and sensitive quantitation of low-abundance neurodegenerative biomarkers in blood. Novel assay panels were developed to target pTau-217, GFAP, NFL, and Amyloid beta 1-42, achieving femtogram-level detectability in healthy plasma samples while maintaining high analytical performance and precision. This work validates a sensitive, analytically robust and automated benchtop solution for the quantitation of neurodegenerative biomarkers in blood, offering a promising tool for advancing biomarker-driven research and clinical applications in neurodegeneration.

The SPEAR UltraDetect™ menu includes key neurological biomarkers and emerging CNS-specific Tau species, improving disease vs. aging discrimination and supporting scalable studies from mechanistic research to clinical applications.
