Species: HumanAssay Type: In VitroCell Line/Tissue: HippocampusWe used ZD-7288 to block Ih current in hippocampal pyramidal cells. 10 µM concentration of the compound developed its effect within a characteristic time interval of tens of seconds, making slower decay kinetics of evoked post-synaptic potentials (see illustration). Washout for 15-20 minutes could not restore the control response characteristics, thus suggesting high affinity of the compound.No problem with dissolution in water and preparation of 10 mM stock.
