Verified Customer | Posted 05/17/2023I have been using Organoid Harvesting Solutions since 2018 for organoid models biochemical analysis.It's very easy to use and allows you to dissociate and depolymerize any basement membrane matrix (Ultimatrix, BME Type 2 Selec, Matrigel) to harvest intact organoids.

Can the Organoid Harvesting Solution, Catalog # 3700-100-01, be used to dissociate cells grown on a substrate of Cultrex Rat Collagen 1?
The Organoid Harvesting Solution works well with BME (Basement Membrane Extract). For cells grown on Cultrex Rat Collagen 1, the recommended enzyme to use to dissociate cells is Collagenase. Collagenase treatment at 37oC for 30 minutes is recommended.

Cultrex Organoid Harvesting Solution (Catalog # 3700-100-01) is a non-enzymatic solution that depolymerizes the matrix to enable harvest of organoids. In contrast, dispase is a protease enzyme that degrades the matrix. End users will need to empirically determine whether enzymatic or non-enzymatic solutions are more appropriate for their experimental setup.
