Specificity

In a functional flow cytometry assay, this antibody binds and blocks SARS-CoV-2 Spike RBD interaction with human ACE-2 transfectants. Detects SARS-CoV-2 Spike RBD and SARS-CoV-2 Spike S1 in sandwich ELISAs when used in the indicated ELISA antibody pairings. Detects SARS-CoV-2 B.1.1.529 S RBD (Omicron Variant) in direct ELISAs.

Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

Background: Spike RBD

The coronavirus Spike protein receptor binding domain (RBD) resides within the S1 subunit and is responsible for binding to host cell receptors and initiating viral infection. Past coronaviruses SARS and MERS along with the global pandemic caused by SARS-CoV-2 have sparked great interest and scientific discovery leading to new vaccines and drug development. At the heart of coronavirus biology lies the Spike protein and its receptor binding domain. The Spike RBD is a 26 kDa domain consisting of a twisted five-stranded antiparallel beta sheet and sits at the apex of each Spike protein monomer. The Spike RBD is flexible thanks to a hinge region that allows for conformational changes that expose (up or open conformation) or hide (down or closed conformation) its receptor contacts. For SARS-CoV-2, the Spike RBD recognizes and tightly binds to the human receptor ACE-2. In the closely related MERS coronavirus, human DPPIV/CD26 acts as the receptor.
