Western blot shows lysates of HEK293 human embryonic kidney cell line, Balb/3T3 mouse embryonic fibroblast cell line, and Nb2-11 rat lymphoma cell line. PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human/Mouse/Rat PLC- gamma1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3288) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for PLC- gamma1 at approximately 155 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

TIM-3 Association with Intracellular Kinases in CD8+/MART-1+ T cells.TIM-3 co-immunoprecipitation analysis of unactivated and 15 min. stimulation with anti-CD3/CD28 beads (activated). Equivalent amounts of protein (~2mg) were co-immunoprecipitated with pAb anti-TIM-3 antibody and western blot was performed using capillary electrophoresis. Cleared lysate served as a loading control for individual antibody reactivity. Background: PLC-gamma 1

Phospholipase C gamma-1 (PLC-gamma 1) is activated by receptor tyrosine kinases in response to growth factors and hormones. Activated PLC-gamma 1 catalyzes the hydrolysis of phosphatidylinositol 4,5-biphosphate to produce the second messengers inositol 1,4,5-triphosphate (IP₃) and diacylglycerol (DAG). IP₃mobilizes the release of calcium while DAG activates protein kinase C. PLC-gamma 1 plays an important role in regulating cell proliferation and differentiation.
