Conjugated Antibodies and Proteins
Conjugated antibodies and proteins enable the visualization of surface and intracellular target proteins, for detection of protein-protein interactions, and for determining the amount of antibody or protein in serum. Whether you are looking for a conjugated antibody for flow cytometry, or a conjugated protein for surface plasmon resonance (SPR) applications, Bio-Techne has the solution for your conjugation needs. Our selection of highly validated, quality antibodies and proteins is constantly growing, and our custom services team is ready to assist if you require a reagent not found in our catalog.
What is Conjugation?
Conjugation, also called labeling or tagging, refers to the process by which a molecule, such as a fluorophore, biotin, or enzyme, is attached to biomolecule, such as an antibody or protein. Conjugated proteins and antibodies are used in a variety of applications including flow cytometry, cytometry by time-of-flight (CyTOF), immunohistochemistry (IHC), immunocytochemistry (ICC), ELISA, and western blot (WB).
Fluorescent tags are fluorophores conjugated to antibodies or proteins. Each fluorophore is excited at a particular wavelength and combining fluorophores with unique emission spectra allows for large multiplex assays. Fluorescent tags are used to detect proteins by flow cytometry, IHC, ICC, and more.

CAR-T Cell Detection by Flow Cytometry
Evaluation of chimeric antigen receptor (CAR) expression following transduction is a critical step in the production of CAR-T and CAR-NK cells for cancer immunotherapy. To simplify CAR detection, R&D SystemsTM fluorescent-labeled proteins bind directly to single chain variable fragment (scFv) CAR T cells. Have confidence in your results when combining fluorescent-labeled recombinant proteins and monoclonal antibodies from Bio-Techne.
Combination of fluorescently labeled antibodies and proteins for detection and immunophenotyping of CAR T cells by flow cytometry. (A) T cells were either transduced with a hCD19-CAR (top) or not transduced (bottom) and then cultured for 11 days. Cells were stained with a PerCP Conjugated Anti-Human CD45 Monoclonal Antibody (Catalog # FAB1430C) and Recombinant Human CD19 Fc Chimera Atto 647N Protein (Catalog # ATM9269). (B) Frequency of transduced cells that are CD4+ or CD8+. Cells were stained with an Alexa FluorTM 405 Conjugated Anti-Human CD8 alpha Monoclonal Antibody (Catalog # FAB1509V) and PE-Cy7 Conjugated Anti-Human CD4.
Biotinylated Antibodies and Proteins
Labeling antibodies and proteins with biotin leverages one of the strongest non-covalent bonds in nature between biotin and streptavidin. Streptavidin has the capacity to bind four biotin molecules, which can dramatically enhance detection of low-abundance proteins. Common applications for biotinylated antibodies and proteins include ELISA, western blot, IHC, SPR, flow cytometry, and more.
For detection of biotinylated proteins and antibodies, Bio-Techne offers:
R&D Systems and Novus Biologicals offer a wide range of monoclonal and recombinant monoclonal biotinylated antibodies for Immune Cell Therapy, Immuno-oncology, and Neurodegenerative Disease research. Find our full catalog of biotinylated antibodies at Novus Biologicals.
Avi-tags: Are short amino acid sequences that allow precise addition of the biotin molecule to a Lysine residue on the Avi-tag. To enhance reliable and reproducible biotinylation results, Bio-Techne offers catalog and custom Avi-tagged biotinylated proteins.
Read our App Note to learn more about the benefits of Avi-tagged biotinylated proteins for protein interaction kinetics and SPR.
Amine- or Sugar-Labeled Biotinylated Proteins: Traditional biotinylation, using amine- or sugar-labeled biotinylation, typically incorporate multiple biotins per molecule, which can be advantageous for increasing signal strength. Similar to Avi-tagged biotinylated proteins, amine- and sugar-labeled biotinylated proteins are tested for lot-to-lot consistency and bioactivity following biotinylation to ensure the activity of the biotinylated protein is the same as the non-biotinylated protein. Enzyme and Peptide Tags
Enzyme Conjugated Antibodies
Bio-Techne offers both primary and secondary antibodies conjugated to HRP and AP. When substrate is added, these enzymes function to induce a color change in colorimetric or chromogenic reactions for ELISA, WB, and IHC detection applications.
Biotechne’s VisUCyte HRP PolymerTM overcomes problems associated with Avidin-Biotin detection chemistry, such as the extra quenching steps required to inhibit endogenous Biotin and Avidin staining.
Peptide Tagged Proteins and Antibodies
Proteins: HIS-tagged proteins are frequency used to aid in purification and protein detection in many different assays. Other tags, such as GST, HA, and Myc can be used for application-specific purposes.
LlaMAbodies: Bio-Techne’s single domain VhH antibodies are smaller than traditional antibodies and can detect hard to reach epitopes for blocking and flow cytometry applications. Single-domain antibodies penetrate fixed tissue more effectively than traditional antibodies and can withstand a greater range of pH and temperature, making them ideal for IHC applications. Conjugation of VhH antibodies to HIS-Tags allows detection of the smallest 15 kDa VhH LlaMAbodies. Bio-Techne has catalog and custom options for single-domain VhH antibodies.
