Background: PP2C gamma

PP2C gamma (Protein phosphatase 2C gamma; also PPM1G) is a nuclear member of the metal ion-dependent PP2C family of Ser/Thr protein phosphatases. Although its predicted MW is 59 kDa, it runs anomalously at 72‑75 kDa in SDS-PAGE. PP2C gamma is widely expressed and serves as a negative regulator of the cell stress response. In particular, it dephosphorylates histones H2A and H2B, allowing for their placement into chromatin. This is important when histones are reincorporated into DNA following DNA repair. Human PP2C gamma is 546 amino acids (aa) in length. It contains a catalytic domain (aa 27‑120), an Asp/Glu-rich region (aa 258‑326) and a poly-Lys tail (aa 539‑544). One potential splice form shows a Phe substitution for aa 20‑38.
