Background: TBX5

TBX5 (T-box protein 5) is a 64 kDa member of the T-box family of transcriptional regulators. It is expressed in both embryo and adult, and is particularly important in the development of the heart and upper limb. TBX5 is a transcriptional activator and influences the expression of FGF‑10 and NPPA/ANF. Human TBX5 is 518 amino acids (aa) in length. It contains one T‑box DNA‑binding domain (aa 58‑238), a transactivation motif (Glu349GluAsp351), and an NLS (aa 339‑379). There are multiple splice forms. One 35 kDa isoform shows a RPLL substitution for aa 252‑518, generating a TBX5 antagonist. Other isoforms show an alternative start site at Met51, a 136 aa substitution for aa 445‑518, a 33 aa substitution for aa 360‑518, a 22 aa substitution for aa 328‑518, and a 41 aa substitution for aa 7‑49. Over aa 253‑327, human TBX5 shares 95% aa identity with mouse TBX5.
