Western Blot: COX-1 Antibody (SC68-05) [NBP2-67643] - Analysis of COX-1 on different lysates with Rabbit anti-COX-1 antibody at 1/1,000 dilution. Lane 1: C2C12 cell lysate Lane 2: A431 cell lysate Lysates/proteins at 10 ug/Lane. Predicted band size: 69 kDa Observed band size: 70 kDa Exposure time: 2 minutes; 8% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Immunohistochemistry-Paraffin: COX-1 Antibody (SC68-05) [NBP2-67643] - Analysis of paraffin-embedded human skin tissue with Rabbit anti-COX-1 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Background: COX-1
Cyclooxygenase (COX), also known as prostaglandin G/H synthase, is a membrane bound enzyme partly responsible for the catalysis of prostanoid synthesis. COX is expressed as at least three different isoforms. COX-1 is constitutively expressed and thought to regulate a number of 'housekeeping' functions such as vascular hemostasis, renal blood flow, and glomerular function. COX-2 expression is tightly regulated and induced by inflammatory mediators such as growth factors, cytokines, and endotoxin. COX-3 appears to be much more strictly regulated spatially and is observed in greatest abundance in cerebral cortex.
