Flow Cytometry: CD161/NK1.1 Antibody (PK-136) - Chimeric - Azide and BSA Free [NBP3-20165] - Paraformaldehyde fixed mouse splenocytes permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-20165 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Western Blot: CD161/NK1.1 Antibody (PK-136) - Chimeric - Azide and BSA Free [NBP3-20165] - RAW264.7(A) (1ug/ml) cell lysate, mouse spleen(B) (1ug/ml) and mouse lymph node(C) (0.3ug/ml) tissue lysate (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with NBP3-20165 before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Background: CD161/NK1.1

NK-1.1 surface antigen is encoded by the NKR-P1B/NKR-P1C gene, also known as CD161b/CD161c and Ly-55. It is expressed on NK cells and NK-T cells in some mouse strains, including C57BL/6, FVB/N, and NZB, but not AKR, BALB/c, CBA/J, C3H, DBA/1, DBA/2, NOD, SJL, and 129. Expression of NKR-P1C antigen has been correlated with lysis of tumor cells in vitro and rejection of bone marrow allografts in vivo. NK-1.1 has also been shown to play a role in NK cell activation, IFN- gamma production, and cytotoxic granule release. NK-1.1 and DX5 are commonly used as mouse NK cell markers. The PK136 antibody has been reported to deplete NK cells, induce redirected lysis, block NK cell function, and induce NK cell proliferation.
