Product Description

Key information: FerroOrange (RhoNox-4) is a highly sensitive and robust fluorogenic probe for the detection of intracellular labile Fe(II). FerroOrange displays weak absorption and low fluorescence in the visible region, but becomes strongly fluorescent upon reaction with Fe(II).

Used for: detection of intracellular labile Fe(II).

Application: high-throughput assays for high content imaging of subcellular labile Fe(II) in live cells; fluorescent microscopy; studies on iron homeostasis and ferroptosis.

Properties and Photophysical Data: FerroOrange has excellent cell-membrane permeability and low cell toxicity. FerroOrange has no chelating ability. For the fluorescent form, excitation and emission maxima (λ) measured in 50 mM HEPES buffer (pH 7.4, 0.2% DMSO) are 545 nm and 575 nm, respectively; extinction coefficient ε = 73,500 M-1cm-1; quantum yield φ = 0.19.

Application of FerroOrange in HeLa cells

Application of FerroOrange in HeLa cells.

HeLa cells treated with chelator of iron 2,2′-bipyridyl (Bpy) (100 μmol/L) or Ammonium iron (II) sulfate (100 μmol/L) were prepared. A: Control; B: Ammonium iron (II) sulfate treated; C: Ammonium iron (II) sulfate and 2,2′-Bipyridyl (Bpy) treated. The change of intracellular Fe2+in HeLa cells was detected by FerroOrange. Excitation/Emision maxima ( lambda) = 561 nm/570-620 nm. Scale bars = 20 μm.
Application of FerroOrange in Hela cells

Application of FerroOrange in Hela cells.

Hela treated with 2 μL of 1 mmol/L FerroOrange and 2 μL of 10 mmol/L from each metal were added to 1 mL of 50 mmol/L HEPES Buffer (pH7.4). The fluorescence intensity was measured after the reaction, for 1hr, at room temperature.
