Product Description
Biotin BromoCatch™ Ligand is a covalent probe designed for the selective labeling and enrichment of proteins tagged with the BromoCatch™ self-labeling tag. Based on a para-acrylamide-functionalized bump-JQ1 scaffold, the BromoCatch™ ligand enables irreversible and highly specific covalent attachment to the engineered cysteine residue in the BromoCatch-tagged protein. The appended biotin handle allows for efficient affinity-based detection, pull-down, and isolation of tagged proteins from complex mixtures such as cell lysates. This probe demonstrates exceptional target selectivity, confirmed via intact protein mass spectrometry and western blotting, and shows no background labeling in untransfected or wild-type samples even at micromolar concentrations. Biotin BromoCatch™ Ligand expands the utility of the BromoCatch™ platform for biochemical workflows including proteomics, immunoprecipitation, and interaction studies.Please refer to this protocol for further information on how to use this product.

Parental or BromoCatch-GFP expressing HEK293 cells were treated with Biotin BromoCatch™ Ligand (1 μM), and probe-interacting proteins were pulled down using streptavidin-coated beads.
