This antibody was prepared by subcutaneously injecting non-polymorphic MHC class II (Ia-like glycoproteins) from the membranes of rat thymocytes into BALB/c mice.

Specificity

This antibody recognises a monomorphic determinant of the rat MHC Class II (I-A) antigen present on B lymphocytes, dendritic cells, some macrophages and certain epithelial cells. It also cross-reacts with mouse MHC, specifically I-A[k] and I-A[s]

Immunocytochemistry/ Immunofluorescence: MHC Class II Antibody (OX-6) - BSA Free [NBP3-43538] - Immunofluorescence analysis of paraformaldehyde fixed rat (Rattus norvegicus) splenocytes on coverslips, permeabilized with 0.15% Triton stained with NBP3-43538 at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 µg/ml), showing membrane and some cytoplasmic staining of a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-43538, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor® 488 secondary antibody.

Western Blot: MHC Class II Antibody (OX-6) - BSA Free [NBP3-43538] - Rat spleen (A), rat thymus (B) and rat lymph node (C) lysate samples (35µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with NBP3-43538 at 1 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for MHC Class II is ~24.5-31.6kDa, depending on the isoform and post-translational modification. NBP3-43538 successfully detected MHC Class II in rat spleen, thymus and lymph node lysates.

Background: MHC Class II

MHC Class II antigens are heterodimers consisting of one alpha chain(31-34kD) and one beta chain (26-29kD). The family of monoclonal antibodies (ER-TR 3, ER-TR 2, ER-TR 1) detect MHC class II antigens encoded by the murine Ia region of the H-2 complex, corresponding to the human HLA-DR region. MHC Class II antigens are a valuable tool for studying T helper cell interaction with class II positive antigen presenting cells (dendritic cells, B cells, macrophages) and offer new possibilities for studying the development of T helper cells since these antibodies also stain stromal cells in the thymus. MHC Class II antigens are also inducible on a number of other cells (endothelium and epithelial cells) by interferon gamma.
