The original antibody was generated by immunizing female ICR mice with human CK20 peptide (amino acids 411-424) conjugated to KLH in equal volume of Freund's incomplete adjuvant. (Uniprot# P35900)
Specificity
This antibody is specific for amino acids 411-424 of human cytokeratin 20. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. There are 19 keratin polypeptides are found in human epithelium. Cytokeratin 20 plays a significant role in maintaining keratin filament organization in intestinal epithelia. When phosphorylated, it plays a role in the secretion of mucin in the small intestine.

Western Blot: Cytokeratin 20 Antibody (10F6B12A9) - Chimeric - BSA Free [NBP3-43567] - Caco-2 (A) cells lysate and human colon (B) tissue lysate (35ug protein in RIPA buffer) were resolved via SDS-PAGE, and the subsequent blots were probed with NBP3-43567 at 1ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and proteins were detected by chemiluminescence.

Immunocytochemistry /Immunofluorescence: Cytokeratin 20 Antibody (10F6B12A9) - Chimeric - BSA Free [NBP3-43567] - Immunofluorescence analysis of paraformaldehyde-fixed Caco-2 cells, permeabilized with 0.15% Triton, on Shi-fix coverslips stained with NBP3-43567 (1:100 dilution) for 1 hour followed by Alexa Fluor 488 secondary antibody (1:1000 dilution), showing cytoplasmic and some nuclear staining. The nuclear stain is DAPI (blue). Panels show, from left to right, top to bottom, NBP3-43567, DAPI, merged channels, and an isotype control.
