Liraglutide inhibits glucolipotoxicity-induced NF kappaB pro-inflammatory responses. (a) Western blots showed that 0.1 μM Liraglutide inhibits glucolipotoxicity-induced NF kappaB p65 subunit phosphorylation at Ser536 and prevents I kappaB alpha degradation at 24 h. (b) Double fluorescence staining revealed that Liraglutide inhibits the entry of the NF kappaB p65 subunit into the nucleus under glucolipotoxicity. Scale bar: 20 μm. (c) qPCR results demonstrated that under glucolipotoxicity, liraglutide significantly reduces the mRNA expression levels of the IL-1 beta, TNF alpha, and MCP-1 pro-inflammatory cytokines, and simultaneously restores the IL-4, IL-10, and IGF-1 anti-inflammatory cytokines. (d) Western blot analysis of NLRP3, ASC, and cleaved-caspase-1 confirmed that Liraglutide suppresses glucolipotoxicity-induced inflammasome activation. Arrows indicate nuclear p65 immunolabeling. All values are presented as the mean +/- SEM. Significant difference was determined using multiple comparisons of Dunnett’s posthoc test for ** p < 0.01 compared to the Control group; ## p < 0.01 compared to the HG+HF group.
