Product Description

Key information: HB-2-30 is a far-red fluorescent transglutaminase-2 (TG2) probe for imaging endocytosis in vitro and in vivo. HB-2-30 undergoes highly efficient endocytosis in complex with TG2 and α2-macroglobulin via the LRP1 pathway.

Used for: Imaging endocytosis in vitro and in vivo, explore targeted lysosomal degradation of secreted and cell surface proteins.

Application: Confocal microscopy.

Properties and Photophysical Data: Suitable for multiplexing and can be combined with DAPI (Cat. No. 5748) counter-staining. Excitation and emission maxima (λ) are 649 and 665 nm, respectively.

Application of HB-2-30 in vitro.

Confocal microscopy images of bone marrow-derived dendritic cells from WT C57BL/6 mice (left) and TG2KO mice (right) incubated with 3μM HB-2-30. Cells were incubated for 90 minutes with the probe before being fixed, stained for nuclei (blue), and imaged. HB-2-30 signals is in red. Application of HB-2-30 in vivo.

Ileum tissue sections from mice dosed with 10 mg/kg HB-2-30. WT C57BL/6 mice (left) and TG2KO mice (right) were dosed with HB-2-30 and sacrificed after 2 hours. Intestines were Swiss rolled, thin sectioned, stained for nuclei (blue), and imaged via confocal fluorescence microscopy. HB-2-30 signal is in red.
