Immunoprecipitation:Asparagine synthetase AntibodyNBP3-29374] -Detection of human ASNS by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-ASNS antibody NBP3-29374 (lot NBP3-29374-1) used for IP at 6 µg per reaction. ASNS was also immunoprecipitated by rabbit anti-ASNS antibody BL21119. For blotting immunoprecipitated ASNS, NBP3-29374 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: Asparagine Synthetase/ASNS

Asparagine synthetase (AS) is a housekeeping enzyme responsible for the production of asparagine from aspartate and glutamine. Most mammalian cells express AS and regulate the level of activity in response to the concentration of asparagine in the medium. Certain tumor cells in contrast, exhibit little or no AS activity and rely on the surrounding medium as a source of exogenous asparagines (1). Therefore tumor cells may be selectively killed by a chemotherapeutic drug using asparaginase. This approach has been exploited in the treatment of certain cancers like childhood acute lymphoblastic leukemia (2).
