Meet SPEAR UltraDetect

Successive Proximity Extension Amplification Reaction (SPEAR) technology overcomes the limitations of conventional proximity extension assays and dramatically reduces transient noise to achieve ultrasensitive and biomarker detection. By integrating nucleic acid amplification into a unique two-factor authentication mechanism for protein detection, SPEAR enables highly specific and robust protein detection in a fully homogeneous, wash-free format using 1 µL of diluted sample.

SPEAR UltraDetect™ Immunoassays employ a simple and scalable workflow, easily adopted by any lab utilizing standard qPCR.

A Homogeneous Assay Without Wash Steps

Step 1: Target Binding

Incubate 1 μL of diluted sample with proprietary SPEAR probes, allowing sufficient time for antibodies to bind to their target.

Step 2: Two Factor Authentication Reaction

Add reaction mix to the well without any washing. Two-factor authentication occurs, the key to achieving unparalleled sensitivity and specificity.

Step 3: Signal Amplification

After the reaction is complete, qPCR mix is added to the well without any washing. After amplification, target-specific signal is quantified from the qPCR results.

What’s more, SPEAR Technology has less stringent requirements on antibody affinity and can consistently achieve high sensitivity across a wide range of biomarkers.

As seen in the figure on the left, SPEAR consistently detected analytes at sub-femtomolar concentrations, achieving LODs 2-6 orders of magnitude lower than the digital ELISA, even when low-affinity antibodies were employed.
