Use of THPTA for clicking fluorophores in a two-step CuAAC mediated reaction.
BromoCatch™ Ligand, Alkyne (Catalog # 8940)(1.5 eq) was pre-incubated with BromoCatch™ or Brd4-BD2WT (purified protein) for 1 h before adding azide sulfonated Cy5 (excess) and the CuAAC reagents (CuSO4 / THTPA/ NaAsc). Samples were taken at 1 and 16 h and analysed by SDS-PAGE. The conjugation selectively labels BromoCatch™, with no labeling observed for the Brd4-BD2WT protein.Product Specifications
