Key information: Rhobo6 ECM Dye is a cell-impermeable fluorescent probe for labeling the extracellular matrix (ECM) in live tissues. It contains a phenylboronic acid group that binds reversibly to diols commonly found in ECM glycans resulting in a significant increase in fluorescence and a red shift in emission spectra. For optimal signal, we recommend using 561 nm excitation and a 575 nm long-pass emission filter. Rhobo6 staining is not compatible with samples that are chemically fixed or that otherwise exhibit compromised cellular membranes.
Used for: labeling the ECM in live tissues.
Application: fluorescent microscopy including 2P and STED microscopy. Enables wash-free visualization of ECM in vitro and in vivo and is suitable for long-term imaging studies.
Properties and Photophysical Data: Rhobo6 provides broad substrate profile, superior tissue penetration, and negligible photobleaching. Excitation and emission maxima (λ) measured in 2 M sorbitol to mimic binding, are 535 nm and 561 nm, respectively; quantum yield = 0.75; extinction coefficient = 97,500 M-1cm-1.
