Precision
Intra-Assay Precision (Precision within an assay) Three samples of known concentration were tested on one plate to assess intra-assay precision.
Inter-Assay Precision (Precision between assays) Three samples of known concentration were tested in separate assays to assess inter-assay precision.

Recovery for Human Thrombospondin-1 Quantikine ELISA Kit
The recovery of Thrombospondin-1 spiked to levels throughout the range of the assay in various matrices was evaluated.
| Sample Type | Average % Recovery | Range % |
|---|---|---|
| Cell Culture Media (n=4) | 95 | 86-101 |
| Platelet-poor EDTA Plasma (n=4) | 94 | 89-99 |
| Platelet-poor Heparin Plasma (n=4) | 93 | 87-104 |
Linearity
To assess the linearity of the assay, samples containing and/or spiked with high concentrations of Thrombospondin-1 were serially diluted with Calibrator Diluent to produce samples with values within the dynamic range of the assay.

Background: Thrombospondin-1
Thrombospondin-1 (TSP-1) is a secreted homotrimeric protein that regulates a wide range of cellular functions including their interactions with other cells and with the extracellular matrix (ECM). TSP-1 counteracts the angiogenic, hypotensive, and anti-thrombotic effects of nitric oxide (NO). It binds and neutralizes VEGF, blocks VEGF R2 signaling on vascular endothelial cells (EC), and destabilizes adhesive contacts between EC. TSP-1 also plays an important role in wound repair and tissue fibrosis by binding latent TGF-beta and inducing release of the active cytokine from the latency associated peptide (LAP). TSP-1 dampens adaptive immune responses by inducing the differentiation of regulatory T cells and inhibiting TCR signaling. In the nervous system, it promotes excitatory synapse formation and supports the integration of neural progenitor cells into the olfactory bulb.
