Immunohistochemistry-Paraffin: SARS-CoV-2 NSP10 Antibody [NBP3-07057] - Immunohistochemical analysis of paraffin-embedded COVID-19 patient lung tissue using anti- SARS-CoV-2 (COVID-19) NSP10 antibody (0.5 ug/mL). Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4 degrees celsius. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin. Strong signal of SARS-COV-2 NSP10 protein was observed in macrophage of COVID-19 patient lung, but not in non-COVID-19 patient lung.

Background: SARS-CoV-2 NSP10

SARS-CoV-2 Nonstructural Protein 10 (NSP10) is one of the sixteen nonstructural proteins of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), the causative agent of COVID-19 (1). SARS-CoV-2 NSP10 is 139 amino acids (aa) with a theoretical molecular weight of 14.8 kDa (1-3). The amino acid sequence alignment of SARS-CoV and SARS-CoV-2's NSP10 has 97.1% sequence identity and 99.3% sequence similarity (1). Functionally, NSP10 interacts with NSP14 which acts as an S-adenosylmethionine (SAM)-dependent (guanine-N7) methyl transferase (N7-MTase) and stimulates it by significantly increasing exoribonuclease activity (1,3,4). Additionally, SARS-CoV-2 NSP10 is proposed to be a cofactor for NSP16, which is a 2'-O-methyltransferase, and the interaction is needed for it to become active (1-4). NSP10 can interact with both NSP14 and NSP16 simultaneously, acting as both a cofactor for each as well as a molecular connector (4). A SARS-CoV-2 interactome study also revealed that NSP10 is involved in vesicle trafficking (2).
