RNF149 downregulates IRF3 protein level through the proteasome pathway.(A) HEK293T cells were transfected with Flag-IRF3 and a gradient concentration of Myc-RNF149 plasmids for 48 h, and the expression of exogenous IRF3 was detected by Western blot. (B) The expression of endogenous IRF3 protein level in HEK293T cells transfected with a gradient concentration of Myc-RNF149 plasmid for 48 h. (C) HEK293T cells were transfected with shRNF149 and Flag-IRF3 plasmids for 48 h, and the expression of exogenous IRF3 was determined by Western blot. (D) The expression of endogenous IRF3 protein level in HEK293T cells transfected with shRNF149 plasmids for 48 h. (E) The expression of IRF3 in peritoneal macrophages from WT and Rnf149−/− mice was detected by Western blot.
