Scientific Data Images for Rat CCL3/MIP-1 alpha Antibody
Detection of Rat CCL3/MIP‑1 alpha by Western Blot.
Western blot shows lysates of NR8383 rat alveolar macrophage cell line untreated (-) or treated (+) with 10 µg/mL LPS for 4 hours. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Rat CCL3/MIP-1a Monoclonal Antibody (Catalog # MAB66252) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for CCL3/MIP-1a at approximately 12 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Chemotaxis Induced by CCL3/MIP‑1 alpha and Neutral-ization by Rat CCL3/ MIP‑1 alpha Antibody.
Recombinant Rat CCL3/MIP-1a (Catalog # 6625-MA) induces chemotaxis in the BaF3 mouse pro-B cell line transfected with human CCR5 in a dose-dependent manner (orange line). Chemotaxis elicited by Recombinant Rat CCL3/MIP-1a (10 ng/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Rat CCL3/MIP-1a Monoclonal Antibody (Catalog # MAB66252). The ND50 is typically 0.075-0.45 µg/mL.Detection of CCL3/MIP-1 alpha by Immunocytochemistry/ Immunofluorescence
Effect of neutralizing antibodies for chemokines/chemoattractans in monocytes/MΦs in vitro cell migration assays. (A) The effect of the neutralizing antibodies against CCL2 (500 ng/mL), CCL3 (500 ng/mL), and CCL21 (500 ng/mL) in the monocytes/MΦs migration assay under the chemoattractant effect of the CM of the glomeruli from DN rats. HAM-F10 and 5% FBS were used as negative and positive control, respectively, of in vitro stimuli for migration. DAPI was used to stain the nuclei of monocytes/MΦs. Representative quadrants show monocytes/MΦs in blue (400× magnification). (B) Quantification of positive intraglomerular area (%) of deconvoluted color for DAB in (A). Graphs represent distribution of each sample and the mean ± S.D. * p < 0.05, and *** p < 0.001 versus Ctrl rats. #p < 0.05 and ###p < 0.001, DN + MRS1754 versus DN rats. n = 4. Application
Recommended Usage
Western Blot
2 µg/mL
Sample: NR8383 rat alveolar macrophage cell line treated with LPS
Sample: NR8383 rat alveolar macrophage cell line treated with LPS
Neutralization
Measured by its ability to neutralize CCL3/MIP‑1 alpha-induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CCR5. The Neutralization Dose (ND50) is typically 0.075-0.45 μg/mL in the presence of 10 ng/mL Recombinant Rat CCL3/MIP‑1 alpha.
