Western Blot: proBDNF Antibody - Azide and BSA Free [NB100-98756]
A thorough Western blot (WB) analysis of tissue lysates and recombinant proteins was performed using NB100-98756 to assess proBDNF expression. The antibody is specific for proBDNF and does not recognize mature BDNF. Specific immunoreactive bands corresponding to proBDNF were detected in both tissue lysates and recombinant protein samples, while no reactivity toward mature BDNF was observed, demonstrating the antibody's specificity for the pro-form of BDNF.Immunohistochemistry-Paraffin: proBDNF Antibody - Azide and BSA Free [NB100-98756]
Immunohistochemistry (IHC-P) on paraffin-embedded rat hippocampus sections detecting proBDNF using NB100-98756. The animal was perfused at a pressure of 130 mmHg with 300 mL of 4% formaldehyde, followed by overnight post-fixation prior to paraffin processing and embedding. Heat-induced epitope retrieval (HIER) was performed in Tris-EDTA buffer (pH 9.0) for 20 minutes. Sections were blocked with 0.2% low-fat dry milk (LFDM) in PBST. The primary antibody was applied at a dilution of 1:1000 and incubated for 30 minutes at room temperature. Detection was performed using a HRP Polymer Detection System, followed by visualization with DAB Chromogen.
