Background: BMP-8b

BMP-8b (Bone morphogenetic protein 8b; also OP-3) is a 16 kDa predicted mature and 45 kDa precurser member of the Gbb-60A class of the BMP family of molecules. In mouse, precursors to sperm and oocytes arise from the proximal epiblast around day E7.5 (days 18-20 in human). This lineage commitment is a response to BMP-4 signaling, which is promoted by the repressive action of BMP-8b on the anterior visceral endoderm, a source of BMP-4 antagonism. Mouse BMP-8b preproprecursor is 399 amino acids (aa) in length. It contains a 19 aa signal sequence, a 241 aa prosegment, and a 139 aa mature region. An interchain disulfide bond occurs at Cys363, generating a homodimer. Over aa 261-399, mouse BMP-8b shares 74% aa identity with mouse BMP-8a, and 69% and 93% aa identity with human and rat BMP-8b, respectively.
