Background: IQGAP1
IQGAP1 (IQ motif with Ras GAP-related domain 1) is a 195 kDa member of the IQGAP family of proteins. It serves as an effector protein for Rho family members, and regulates actin polymerization. Human IQGAP1 is 1657 amino acids (aa) in length. It contains one actin-binding CH domain (aa 45‑159), a Pro-binding WWP region (aa 685‑711), four consecutive calmodulin-binding IQ motifs (aa 745‑864), and a noncatalytic Ras-GAP domain (aa 1004‑1237). Amino acids 763‑863 mediate homodimerization. There are at least three IQGAP1 splice variants. One shows a two Lys substitution for aa 270‑1657, a second shows a ten Lys substitution for aa 299‑1657, and a third shows a three Lys substitution for aa 941‑1657.
