Detection of Human CLOCK by Western Blot.
Western blot shows lysates of PC-3 human prostate cancer cell line and HUVEC human umbilical vein endothelial cells. PVDF membrane was probed with 0.5 µg/mL of Human CLOCK Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4026) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for CLOCK at approximately 95 kDa (as indicated). This experiment was conducted using Immunoblot Buffer Group 1.Applications for Human CLOCK Antibody

Background: CLOCK
CLOCK is an essential regulator of circadian rhythms. CLOCK is a basic helix-loop-helix-PAS transcription factor that heterodimerizes with BMAL1. Together these transcription factors bind to E box elements (CACGTG) within the promoter of circadian genes (period1 and 2) stimulating their transcription. BMAL dimerization is necessary for CLOCK’s nuclear translocation, E box dependent transcription, phosphorylation, and degradation.
